Seems like Molly, Carey, Clu, Jack, Fi, and Annie all play the guitar. At least I think I remember Clu with one, but I can't recall what ep. IIRC, Fi got an instructional comment from Molly once and another time said she only knew a few chords? Anyway, not that proficient, I guess. Molly also plays piano, mentioned learning from dad in "The Rock" and she played in "Oopa." plays the harmonica. Anyone else got any others to contribute?

Carey plays the harmonica too in season 3.

It's mentioned that Annie can play almost any instrument. She's only ever really seen playing the guitar, but in Lightning Rod, her mother says "She can pick up almost any instrument and play it, it comes naturally to her."

Annie also plays the flute she made out of a stick in Detention.

Not sure if the instructional comment you mentioned was about Fi and Molly in Medium, but it reminded me of the end of Tulpa when Fi plays She Sells with Jack and Molly. They each had a guitar and I guessed Fi's was her own. Now I'm curious if she was borrowing from someone.
Clu had a guitar in Family Reunion! It's in his very first scene. It's electric, but he has an acoustic in Simplicity when they sing together at the end.
Coincidentally, Jack's first appearance is also with a guitar, and in Simplicity we see him with it again (when they're teasing Molly about the jingle). It's mentioned that Annie can play almost any instrument. She's only ever really seen playing the guitar, but in Lightning Rod, her mother says "She can pick up almost any instrument and play it, it comes naturally to her." Sounds a lot like Jennifer in Rewind. I guess the s3 writers were inspired by their own words.
